多光子蛍光顕微鏡はサブミクロンスケールでの生体組織の三次元非侵襲的研究を可能にする生物学的イメージングの重要な技術となっています。多光子顕微鏡法におけるコントラストのメカニズムは、2つ以上の光子による蛍光体の励起に基づいています。観察標本において励起された蛍光体は検出された可視光の光子を放出し、再び緩和状態に至ります。分子が可視域の単一光子で励起される従来の線形蛍光顕微鏡とは対照的に多光子顕微鏡における特徴的な非線形特性と励起波長は、いくつかの利点を観察者に提供します。(i) 赤外光子の吸収が減少することでより大きなプロービング深度が得られ、また観察標本への吸収損傷が少ないためインビボでのイメージングが可能になる。(ii) 蛍光励起は顕微鏡の焦点面に限定されているため、空間的なフィルタリングが不要である。等の利点が上げられます。
